High Acrylamide Percentage for Target Protein Size

Symptom
Higher molecular weight proteins remain near the loading wells with minimal migration, resulting in poor resolution and bands clustered at the top of the membrane.
Common Causes
  1. 1 Acrylamide concentration too high for large molecular weight proteins, creating excessive gel matrix density
  2. 2 Gel pore size too small for target protein (>100 kDa), physically hindering migration
  3. 3 Incorrect gel recipe selection without considering protein molecular weight range
  4. 4 High acrylamide density impeding effective protein transfer during electroblotting
Solutions
  1. 1 Use 6-10% acrylamide gels for proteins larger than 100 kDa
  2. 2 Consult vendor gel recipe guides to match acrylamide percentage with target protein molecular weight
  3. 3 Reduce acrylamide concentration in custom gel preparations according to validated protocols
  4. 4 Consider using gradient gels (4-20%) when analyzing samples with wide molecular weight ranges
