Incomplete cellular fixation due to insufficient incubation

Symptom
Cells show incomplete crosslinking when fixation time is inadequate, resulting in poor preservation, continued enzymatic activity, or inadequate biosafety inactivation of infectious samples. Fixed cells demonstrate degradation during storage or analysis.
Common Causes
  1. 1 Fixation with 1% PFA requires 45-60 minutes for complete cellular crosslinking
  2. 2 4% PFA concentration requires minimum 15-20 minutes incubation for full fixation
  3. 3 Insufficient fixation time fails to inactivate HIV and other pathogens to biosafety standards
  4. 4 Premature termination of fixation leaves enzymatic activity (phosphatases, proteases) partially active
Solutions
  1. 1 Incubate samples for 45-60 minutes when using 1% PFA fixative solution
  2. 2 Incubate samples for minimum 15-20 minutes when using 4% PFA (e.g. BioLegend buffer)
  3. 3 Use minimum 0.37% PFA with adequate incubation time for HIV-infected sample disinfection
  4. 4 Verify complete fixation by checking for cessation of morphological changes under microscopy
  5. 5 Allow extra fixation time (up to 60 minutes with 4% PFA) for critical biosafety applications
