No Band or Faint Band Due to Reagent Concentration Issues

Symptom
Gel shows no amplification product or very weak bands despite correct thermal cycling parameters, indicating problems with reaction component concentrations.
Common Causes
  1. 1 dNTP concentration incorrect (too high causes Mg2+ depletion; too low limits synthesis; should be 200 µM each)
  2. 2 Primer concentration incorrect (too high >1 µM causes nonspecific binding; too low <0.2 µM causes inefficient annealing)
  3. 3 Insufficient Mg2+ (not enough or omitted; should be 1.5 mM final concentration)
  4. 4 Enzyme concentration too low (insufficient polymerase for complete replication)
  5. 5 Insufficient template (too low initial amount requires increased cycles or more template)
  6. 6 Impure reagents (contaminated dNTPs, primers, or water inhibit PCR)
Solutions
  1. 1 Use 200 µM of each dNTP in final reaction
  2. 2 Use well-designed primers at 0.2–1 µM final concentration; verify manufacturer concentration
  3. 3 Use 1.5 mM Mg2+ in final reaction; verify not omitted
  4. 4 Use optimal enzyme concentration based on template length and difficulty
  5. 5 Increase template amount or increase amplification cycles in increments of 5
  6. 6 Use high-quality dNTPs, desalted or highly purified primers, and fresh nuclease-free water
