Suboptimal Antigen Retrieval Method Selection

Symptom
Weak or absent staining despite proper antibody concentration and incubation. Clear performance differences observed when comparing different antigen retrieval heating methods.
Common Causes
  1. 1 Water bath used for antigen unmasking (not recommended method)
  2. 2 Heating method incompatible with specific tissue type or antigen target
  3. 3 Antigen unmasking buffer not optimized for specific tissue or target protein
  4. 4 Using old or improperly prepared 1X antigen retrieval solutions instead of fresh daily preparations
Solutions
  1. 1 Use microwave oven for antigen retrieval (preferred method over water bath)
  2. 2 For certain tissues/antigens requiring enhanced signal, use pressure cooker instead of microwave
  3. 3 Follow CST product-specific datasheet for recommended antigen unmasking buffer and protocol
  4. 4 Prepare fresh 1X antigen retrieval solutions daily; do not reuse or store diluted buffers
