Low cfDNA Yield Due to Improper Reagent Handling

Symptom
Cell-free DNA extraction yields are lower than expected. This may manifest as insufficient DNA concentration measured by fluorometry or qPCR, preventing downstream analysis.
Common Causes
  1. 1 Monarch Mag Beads M2 used directly from 4°C storage without warming to room temperature
  2. 2 Buffers improperly prepared with incorrect concentrations or volumes
  3. 3 Incorrect buffer added at specific protocol steps
  4. 4 Reagents stored at improper temperatures affecting performance
Solutions
  1. 1 Allow Monarch Mag Beads M2 to warm to room temperature before use
  2. 2 Verify buffer preparation according to protocol specifications and confirm correct volumes
  3. 3 Cross-check buffer addition at each step against the protocol
  4. 4 Store all components at appropriate conditions (Beads at 4°C, verify other reagent storage requirements)
