Low DNA Yield from Gel Extraction or PCR Cleanup Due to Incomplete Elution

Symptom
DNA recovery from gel extraction or PCR cleanup is lower than expected based on input amount. Some DNA likely remains bound to column.
Common Causes
  1. 1 Elution buffer not delivered directly to center of column membrane
  2. 2 Insufficient elution volume or incubation time for complete DNA release
  3. 3 DNA fragments >10 kb eluted at room temperature instead of 50°C
  4. 4 Single elution used when multiple rounds could improve recovery
Solutions
  1. 1 Deliver Elution Buffer directly to center of column membrane, not sides
  2. 2 Use larger elution volumes and extend incubation time to increase yield
  3. 3 For DNA >10 kb, heat DNA Elution Buffer to 50°C and extend incubation to 5 minutes
  4. 4 Perform multiple rounds of elution to maximize DNA recovery from column
