Chemical Contamination from Residues

Symptom
Poor cell growth, cell death, or altered cell behavior without visible microbial contamination. May include effects from free radicals, metal ions, disinfectant/detergent residues, or endotoxins persisting after bacterial contamination is cleared.
Common Causes
  1. 1 Use of non-laboratory-grade water for preparing buffers and solutions
  2. 2 Inadequate rinsing of reusable labware leaving detergent residue
  3. 3 Autoclaving does not remove detergent residue from glassware
  4. 4 Endotoxins present in media, supplements, or serum without certification
  5. 5 Contaminated reagents or water used in media preparation
  6. 6 Equipment and supplies releasing chemical contaminants
Solutions
  1. 1 Always use laboratory-grade water for preparing buffers, solutions, and resuspending lyophilized reagents
  2. 2 Rinse all reusable labware exhaustively and allow to air-dry completely
  3. 3 Remember that autoclaving has no effect on detergent residue removal
  4. 4 Obtain media, supplements, and serum exclusively from suppliers providing endotoxin testing certification
  5. 5 Use high-quality, certified reagents from reputable suppliers
  6. 6 Implement quality control testing for chemical contaminants in prepared media
