Cell Culture (Cell Death) severe

Cell Death from Poor Quality or Toxic Reagents

Symptom
Sudden cell death occurs immediately after introducing new lot of media, serum, trypsin, or buffer. Previously healthy cultures show rapid necrosis or apoptosis within 12-24 hours.
Common Causes
  1. 1 Endotoxin contamination in media or serum exceeding 1 EU/mL causing inflammatory cell death
  2. 2 Heavy metal contamination (copper, zinc) from improper water source or expired media components
  3. 3 Incorrect osmolarity from formulation errors: physiological range 260-320 mOsm/kg, deviations >10% toxic
  4. 4 Expired or improperly stored reagents with degraded growth factors, vitamins, or amino acids
  5. 5 Residual detergent or sterilant contamination from improperly rinsed lab glassware or filters
Solutions
  1. 1 Implement lot testing protocol: test all new reagent lots in parallel with proven lots for 3-5 days before full adoption
  2. 2 Request certificates of analysis (CoA) for all critical reagents; verify endotoxin levels <0.5 EU/mL
  3. 3 Measure osmolarity of prepared media using osmometer; adjust to 290±20 mOsm/kg before use
  4. 4 Store media at 2-8 °C protected from light; use within 2 months of opening or per manufacturer guidelines
  5. 5 Use only tissue culture-grade water (Type I, 18.2 MΩ·cm) and disposable sterile plasticware to avoid contamination
  6. 6 Maintain parallel control cultures with previous reagent lots during changeover period
