Temperature-Induced Protein Precipitation in Culture Media

Symptom
Turbidity or visible precipitates appear in cell culture media after temperature changes. High-molecular weight plasma proteins fall out of solution, creating visible cloudiness that may interfere with microscopy and imaging assays.
Common Causes
  1. 1 Extreme temperature shifts causing protein denaturation
  2. 2 Heat inactivation procedures promoting protein aggregation
  3. 3 Repeated freeze/thaw cycles of serum or media components
  4. 4 Taking refrigerated media directly to 37°C incubator without equilibration
Solutions
  1. 1 Follow manufacturer's guidelines for optimal storage and handling temperatures
  2. 2 Avoid repeated freeze/thaw cycles; aliquot serum and supplements before freezing
  3. 3 Allow refrigerated media to warm gradually to room temperature before use
  4. 4 Use single-use aliquots for heat-sensitive components
