High Background with Nonspecific Bands

Symptom
Western blot shows high background signal with multiple nonspecific bands, obscuring the target protein band and making interpretation difficult.
Common Causes
  1. 1 Non-specific protein binding to agarose beads during immunoprecipitation
  2. 2 Beads not properly blocked with BSA prior to antibody addition
  3. 3 Excessive bead volume used in the IP procedure
  4. 4 Wash buffer stringency too low to remove nonspecific binding
  5. 5 Non-specific antibody binding due to excessive incubation time, lysate concentration, or antibody concentration
  6. 6 Inadequate number or duration of washing steps
Solutions
  1. 1 Perform pre-clearing step using non-coated agarose beads prior to adding antibody-coated beads to remove proteins that bind nonspecifically
  2. 2 Ensure beads are properly blocked with BSA before antibody addition
  3. 3 Decrease the volume of beads used in the protocol
  4. 4 Switch to higher stringency wash buffer such as RIPA-based buffer
  5. 5 Decrease incubation time, lysate volume/concentration, and/or antibody concentration
  6. 6 Add more washes and/or extend wash duration; invert tube several times during each wash to ensure thorough mixing
