Weak or Faint Bands in Western Blot

Symptom
Protein bands appear faint or barely visible on the Western blot membrane or film despite proper loading. Signal intensity is insufficient for detection or quantification.
Common Causes
  1. 1 Blocking reagent has affinity for target protein, masking epitopes
  2. 2 Antibody concentration too low or antibody degraded by multiple freeze-thaw cycles
  3. 3 Insufficient antibody incubation time
  4. 4 Detection reagent outdated or HRP inhibited by sodium azide in buffer
  5. 5 Using monoclonal antibody instead of polyclonal (fewer epitopes)
  6. 6 Chromogenic detection substrate inactivated by tap water instead of Milli-Q water
Solutions
  1. 1 Switch blocking reagent (e.g., from BSA to casein) or reduce blocking time/concentration
  2. 2 Increase primary and secondary antibody concentration; prepare fresh aliquots; protect fluorescent antibodies from light
  3. 3 Extend antibody incubation time (overnight at 4°C for primary antibody)
  4. 4 Use fresh chemiluminescent substrate; eliminate sodium azide from all buffers
  5. 5 Switch to polyclonal primary antibody for multiple epitope recognition, or use biotin-conjugated secondary antibody for signal amplification
  6. 6 Prepare all chromogenic reagents with Milli-Q water; rewet dried chromogenic blots in water
