Speckled or Splotchy Membrane Background

Symptom
Membrane shows irregular spots, speckles, or splotches distributed across the surface. Background is uneven with localized dark regions rather than uniform signal.
Common Causes
  1. 1 Contaminated transfer sponges harboring bacterial growth from previous use without proper sanitization
  2. 2 Contaminated buffers: buffers older than a few days contain bacterial and fungal contaminants
  3. 3 Reused diluted antibody prone to microbial contamination after storage
Solutions
  1. 1 Use new transfer sponges or thoroughly sanitize old sponges with 10% bleach solution followed by extensive rinsing
  2. 2 Always prepare fresh buffers for each experiment; discard buffers after use or within 1-2 days
  3. 3 Use freshly diluted antibody for each experiment; never store and reuse diluted antibody solutions
  4. 4 Filter buffers through 0.22 µm filters if contamination is suspected
