PCR Tube Crushing or Deformation Under Lid Pressure

Symptom
PCR tubes become crushed, collapsed, or deformed after thermal cycling, potentially causing sample loss or compromised seal integrity. Tubes may show visible damage especially when using tube strips.
Common Causes
  1. 1 Incompatible cap style or tube design not verified for specific thermal cycler model
  2. 2 Excessive pressure from thermal cycler lid due to overtightening or malfunctioning spring mechanism
  3. 3 Uneven distribution of tubes/strips across block creating pressure imbalance under lid
Solutions
  1. 1 Use PCR plastics selection tool to verify tube and cap style compatibility with thermal cycler manufacturer specifications
  2. 2 Distribute tubes and strips evenly across block; place empty tubes if needed to balance lid pressure
