Large Error Bars in Quantitative Western Blotting

Symptom
Quantitative measurements show high variability between replicates. Large error bars or standard deviations prevent statistical significance despite apparent differences.
Common Causes
  1. 1 Inconsistent Western blotting conditions between experimental runs
  2. 2 Different antibody lots used across experiment replicates
  3. 3 Variability in transfer efficiency, blocking, washing, or detection steps
  4. 4 Insufficient number of biological or technical replicates
Solutions
  1. 1 Standardize all Western blot conditions: use identical reagent concentrations, incubation times, and temperatures across all experimental runs
  2. 2 Use same antibody lot numbers for entire experiment; purchase sufficient quantity or reserve aliquots from single lot
  3. 3 Process all samples in parallel when possible; include same control samples on every blot for normalization
  4. 4 Increase number of biological replicates (n≥3) and technical replicates; perform power analysis to determine required sample size
