High Non-Specific Background Signal in Competitive ELISA

Symptom
Elevated background signal is detected across wells, interfering with specific signal measurement. Background noise reduces assay sensitivity and may mask low-concentration analyte detection.
Common Causes
  1. 1 Detector conjugate binding non-specifically to the microplate surface
  2. 2 Excessive concentration of detector conjugate antibody
  3. 3 Inadequate blocking buffer composition or insufficient blocking incubation
  4. 4 Insufficient washing cycles or inadequate wash duration
Solutions
  1. 1 Run control wells with primary antibody and sample omitted to identify non-specific conjugate binding
  2. 2 Titrate and decrease the concentration of detector conjugate antibody
  3. 3 Optimize blocking by testing alternative blocking buffers or extending blocking time/temperature
  4. 4 Increase the number of wash cycles and/or extend wash duration per cycle
