Incomplete Standard Reconstitution with Visible Particulates

Symptom
After adding reconstitution buffer to lyophilized standard, visible undissolved material or particulates remain in the vial, leading to inaccurate standard concentrations.
Common Causes
  1. 1 Insufficient mixing time or gentle agitation after adding reconstitution buffer
  2. 2 Protein aggregation due to rapid addition of buffer or vortexing at high speed
  3. 3 Standard vial not centrifuged before opening, causing lyophilized material to adhere to cap or walls
  4. 4 Incorrect reconstitution buffer used or buffer at inappropriate temperature
Solutions
  1. 1 Briefly centrifuge the standard vial (pulse spin) before opening to collect lyophilized material at bottom
  2. 2 Add reconstitution buffer slowly down the side of the vial to avoid foam formation
  3. 3 Mix by gentle swirling or inversion for 5-10 minutes; avoid vigorous vortexing which can denature protein
  4. 4 Visually inspect vial against light after reconstitution to confirm complete dissolution
  5. 5 Allow reconstitution buffer to equilibrate to room temperature before use
  6. 6 If particulates persist, centrifuge briefly and use only the clear supernatant for standard preparation
