Incubation Temperature Below Optimal Range

Symptom
Consistently weak signal across all samples including positive controls. Signal improves when experiment is repeated with attention to temperature. Longer incubations partially compensate.
Common Causes
  1. 1 Reagents and plates not equilibrated to room temperature before use (e.g., used directly from 4°C storage)
  2. 2 Incubation performed in cold room or on cold bench instead of at specified temperature
  3. 3 Room temperature lower than recommended range (typically 20-25°C required)
  4. 4 Cold reagents reduce antibody-antigen binding kinetics and enzyme activity
Solutions
  1. 1 Equilibrate all reagents and coated plates to room temperature (20-25°C) for 30 minutes before use
  2. 2 Perform all incubation steps at manufacturer-specified temperature (typically room temperature or 37°C)
  3. 3 Use temperature-controlled incubator or water bath for precise temperature control during incubations
  4. 4 Monitor actual incubation temperature with thermometer; avoid drafty areas or direct sunlight that affect temperature
